Theodoros Stamos wird an Silvester 1922 als Sohn griechischer Einwanderer in New York geboren. 1936 beginnt er an der Peter Stuyvesant Highschool mit einem Kunststudium, erhält später auch ein Stipendium von der American Artist’s School, um sein Talent als Bildhauer weiterverfolgen zu können. Im Jahr darauf trifft Stamos auf Joseph Solman von der Künstlergruppe "The Ten", welcher ihm wiederum die Malerei näherbringt und für Stamos eine Art Mentor wird. Auch erkennt er, dass der Aufwand bei bildhauerischen Arbeiten um einiges grösser ist – allein für die Materialkosten und Mieten kann Stamos nicht aufkommen. So hält er sich als Gast in verschiedenen Ateliers rund um die 10th Street auf, während deren Zeitspanne die ersten Bilder von amerikanischen und griechischen, schlicht gehaltenen Landschaften entstehen. 

 Auf Solmans Drängen hin besucht Stamos die einflussreiche Art American Place Gallery von Alfred Stieglitz, wo er Arbeiten von Arthur Dove und Georgia O'Keeffe sieht. Während dieser Zeit, in den späten 1930er und frühen 1940er Jahren, hat der junge Künstler eine Vielzahl von Gelegenheitsjobs inne: Drucker, Florist, Hutmacher und Buchverkäufer. Durch einen Job, in Herbert Benevys Rahmengeschäft Gramercy Art in der East 18th Street, lernt er Mitglieder der europäischen Avantgarde kennen, darunter Arshile Gorky und Fernand Léger. 

 1943 hat der erst 21-Jährige seine erste Solo-Ausstellung in Betty Parsons Wakefield Gallery, sie bleibt bis 1957 eine wichtige Bezugsperson für Stamos Künstlerkarriere. Ausstellungen im Whitney Museum (1945 – 1951), Carnegie Institute und Art Institute of Chicago (1947) und im MoMA (1948) folgen. 

 Die während der 1940er entstehenden Werke kombinieren gedämpfte Farben mit biomorphen Motiven, die geologische oder organische Formen suggerieren, passend zu Stamos Interesse für die Naturwissenschaft. In den späten 40ern wird er Teil der Irascible Eighteen, einem Zusammenschluss von Malern, die gegen die Politik des MET protestiert haben und als die erste Generation der amerikanischen abstrakten Expressionisten angesehen werden: rund um Willem de Kooning, Adolph Gottlieb, Hedda Sterne, Jackson Pollock, Clyfford Still, Robert Motherwell, Mark Rothko und anderen bildet Stamos fortan einen Teil der New York School. 

 In den späten 1950ern greift er zunehmend auf reduziertere und vereinfachte Kompositionen zurück; verwendet hierbei dünne Pigmentschichten, um in seinen breiten Farbflächen Tiefe zu generieren. Beispielhaft hierfür sind Bilder der Infinity Series aus den 70ern oder Stamos Sun Boxes; mehrere lange Serien von Gemälden, die ab 1962 entstehen. Die Abstraktionen dieser Reihen zeichnen sich durch breite Farbflächen aus, die durch schlanke Linien oder Formen abgegrenzt werden. 

 Von 1950 bis 1954 unterrichtet er am Black Mountain College und von 1955 bis 1975 lehrt er an der Art Students League of New York und der Cummington School of Fine Arts. Stamos ist ausserdem Mitglied der Uptown Group. Ein Jahr vor seinem Tod schenkt er 43 seiner Werke der Nationalgalerie Griechenlands. Theodoros Stamos stirbt am 2. Februar 1997.
